Methods for estimating number, length, surface area and volume of microscopical structures, based on evaluation of three-dimensional image data obtained by confocal microscopy, are presented: (i) interactive unbiased stereological methods using virtual spatial grids generated by a computer applied to thick slices of arbitrary orientation; (ii) automatic digital methods based on image processing; (iii) automatic digital methods based on surface triangulation. The application of all methods, using our programs and software modules, is shown on practical examples. The methods are compared from the point of view of their applicability, efficiency and precision.
