Identification and isolation of boar sperm specific antigens with potential role in sperm-egg interaction.

Investigations on specific and functionally active sperm antigens would bring about the elucidation of the mechanisms of gamete interaction and help the search to new approaches for prognosis and regulation of fertility.We used polyclonal rabbit anti-boar spermatozoa antibody (RABSA)by double immunodiffusion, electrophoresis, immunofluorescence test and ELISA. The RABSA stained acrosomes. The RABSA is specifically localized in sperm structures. In WB RABSA recognized specifically antigen with molecular mass 60 kDa (Ag60). The biological and IIF experiments provide evidence supporting the involvement of Ag60 in functional steps required for sperm-egg binding and/or fusion, but not sperm-zona pellucida binding

Studium specifických a funkčně aktivních antigenů spermií by mohlo napomoci objasnit mechanismy interakce gamet a najít nové postupu prognózy a regulace plodnosti. Využili jsme polyklonální králičí protikančí protilátku (RABSA) k dvojité imunodifúzi, elektroforéze, imunofluorescenčnímu testu a testu ELISA. Protilátka RABSA barvila akrozomy. RABSA je specificky lokalizován ve spermatických strukturách. U Westernova přenosu rozpoznávala RABSA specificky antigen s molekulovou hmotností 60 kDa (Ag60). Biologické a IIF experimenty prokazují, že Ag60 se účastní funkčních kroků nezbytných pro vazbu spermie na vajíčko a/nebo jejich fúzi, ale ne pro vazbu spermie na zonu pellucidu
