Porcine beta-microseminoprotein (MSP) has been described as a protein characteristic of the prostate; however, we proved its mRNA transcripts in all reproductive organs. In tissue extracts, specific antibody antiMSP recognized a 12-kDa protein band, which was identified by mass spectrometry as MSP, and immunofluorescence displayed the MSP occurrence in epithelial tissue. MSP was localized on epididymal sperm in the acrosomal region and in addition to that on the flagellum of ejaculated sperm. The MSP absence on the surface of capacitated sperm together with the antibody detection of MSP in the sperm acidic extract after in vitro capacitation indicated its acrosomal origin. The MSP sperm localization was confirmed by electron microscopy. The secretion of porcine MSP by different reproductive organs might indicate its multiple roles in the reproductive process, and for the first time in mammalian species, the MSP localization has been shown at different physiological stages of sperm.
