The non transcribed spacers of both 5S rDNA families from Nicotiana tabacum as well as from corresponding diploid parents were cloned. Their sequences differ mainly in the central part where N. tomentosiformis like spacer contains 141 bp long, CG rich insertion. No detectable changes occurred during evolution of individual families in tobacco allotetraploid nucleus indicating relatively recent origin of N. tabacum or low recombinant activity between both parental 5S rDNA gene families.
