Human embryonic stem cells (hESCs) are derived from the inner cell mass of the blastocyst stage embryo and they have ability to differentiate into any cell type. In our experimental system, changes in the chromatin structure were studied in hESCs undergoing differentiation induced by all-trans retinoic acid (ATRA). Nuclear patterns of the Oct4 (6p21.33) gene, responsible for hESC pluripotency and the C-myc (8q24.21) gene, that controls cell cycle progression, were analyzed.
Lidské embryonální kmenové buňky mají schopnost dát vznik kterékoliv buňce lidského těla. Diferenciační děje, které souvisí se změnou multipotentní kmenové buňky na specializovaný typ podmiňují také její změny struktury chromatinu. V našem in vitro experimentálním systému, změny chromatinové struktury, definované genem Oct4 (6p21.33), jsme studovali v průběhu diferenciace lidských embryonálních buněk indukovaný kyselinou retinovou.
