We synthesized and tested phosphonate oligonucleotide of various length mimicking the processed U5 terminus of the proviral HIV-1 LTR for strand transfer inhibition. To study a proces of assembly of immature viral particles and possibly its inhibition, we have identified the minimal Gag assembly domains, which are necessary for formation of virus-like particles in vitro. Two models were used for this study, HIV-1 and Mason-Pfizer monkey virus, a representative of betaretroviruses, which assembles immature particles within the host cytoplasm.
Byly syntetizovány a testovány fosfonátové oligonukleotidy různé délky, které byly odvozeny od HIV-1 LTR, pro inhibici strand transferové aktivity HIV-1 integrasy. Pro studium inhibice sbalování nezralých virových částic jsme identifikovali minimální doménu, která za in vitro podmínek tvoří virové částice. Pro studium jsme použili jako model M-PMV a HIV-1.
