    AnnotationA previously unknown gene, FAM208A, has now featured predominantly in several different genome-wide screens for modifiers of gene silencing. It is now known that FAM208A exists within a core epigenetic silencing complex comprising also MPHOSPH8 and PPHLN1. This complex, termed HUSH (human silencing hub), binds pre-existing, repressive H3K9me3 marks and then catalyses the local spread of these same marks through the recruitment of the H3K9-methyltransferase, SETDB1. Here we discuss the role of forward genetic screens in the discovery of FAM208A functions and the prospects of employing forward chemical genetics screens to further probe its mechanism of action.
