We have developed an ESEM for in-situ observation of live nature. Our AQUASEM II is base on a Tescan VEGA SEM. Its exceptionality is an original differentially-pumped chamber, which can be evacuated slowly and gently to the sample, and a specific detection system with four detectors. On the basis of our new methodology proved by the results of Monte Carlo simulations of beam electrons scattering in water, we have recorded the moving of a Stored-product mite alive for ~5 min at a vapor pressure of 950 Pa at 7oC. Our results demonstrate that this microscope is well applicable to in-situ observations of natural structure of biological or organic samples in wet modes, non-coated surfaces of materials such as semiconductors and oxides, growth or reaction processes of nanoparticles in solution, etc.
