The signal of the unreacted label is routinely significantly higher, indicating the need to deplete the unreacted label prior to analysis by CE. The suitability of five different solid-phase extraction sorbents (Spe-ed Amide-2, Chromabond HILIC, HyperSep Diol, Bond Elut PBA, and HyperSep Hypercarb) was tested for the clean-up and concentration of positively labeled maltooligosaccharides from the reaction mixture. Maltooligosaccharides containing four to seven glucose units were labeled by cationic tags (2-aminoethyl)trimethylammonium chloride and (carboxymethyl)trimethylammonium chloride hydrazide (Girard’s reagent T). Purified samples were analyzed by CE with conductivity detection to compare the purification selectivity and efficiency using the tested SPE materials. The amide-based sorbent gave the best results and was used to process the N-linked glycans enzymatically released from the ribonuclease B sample.
