    AnnotationWe proposed a PDMS droplet-based microfluidic chip, with passive cell encapsulation, passive mixing, and incubation part. Discrete droplets with at least one cell per droplet in a regulated fashion of about 1Hz were formed and the cell-lysis concept using detergent Triton X-100 was optimized. The examined approach has essential benefits for cell manipulation, separation, and analysis of the droplet content.
