Explant cultures treated with pharmaceutical inhibitors started at E12.5 and E13.5, respectively, up to 72 h. Apoptosis was evaluated by appearance of apoptotic bodies and TUNEL test, proliferation by PCNA staining in serial histological sections. General inhibition of caspases in tooth germ cultures led to lack of apoptotic bodies and specific DNA fragmentation, caspase-3 inhibition caused significant decrease of apoptotic cells after 24 h inhibition. PCNA staining did not detect any differences between experimental and control groups.
