Diabetes represents a major complication for pregnancy because it increases the risk for congenital abnormalities in humans. Diabetic embryopathy can affect any developing organ system, although cardiovascular malformations and neural tube defects are the most prominent congenital malformations associated with diabetic pregnancy.Although the development of congenital malformations associated with maternal diabetes is well recognized, it is not well understood. Using the mouse as an experimental system and gene expression profiling, we have identified genes whose expression is significantly changed in embryos exposed to maternal diabetes. A substantial fraction of these genes is known to be essential for normal embryonic development and metabolism. Interestingly, the largest number of genes (25/146) was in the transcriptional factor functional category. We hypothesize that the metabolic imbalance occurring in diabetic pregnancy alters transcriptional regulation in the developing embryo
