Micronucleus assay, which has been used for almost sixty years, is one of the basic methodological approaches of genetic toxicology. This is also evident from more than 10,500 references found using the keyword “micronucleus” in PubMed database. These studies, both in vitro and in vivo, which are discussed in the context of deleteriousness for humans, concern the evaluation of cytotoxicity and genotoxicity of numerous chemicals and in the last decade also nanomaterials (NMs) and nanoparticles (NPs). The aim of this mini-review was to track down the story of the development, improvement and possibilities of the micronucleus assay and revise its significant achievements in the field of nanotoxicology research in both in vitro and in vivo studies.
