Celý postup spočívá v následujících krocích: izolace a imobilizace NF-M, příprava sloučenin k depleci oligonukleoticů z knihovny oligonukleotidů, které by se mohly nespecificky vázat na NF-M, příprava oligonukleotidové knihovny pro screening, výběr a amplifikace oligonukleotidů vázajících se na NF-M, vazebné testy se značenými oligonukleotidy, klonování a sekvenování vybraných oligonukleotidů a funkční charakterizace syntetických aptamerů. Description in English Technology of selection and preparation of aptamer to neurofilament protein NF-M describes individual steps in preparation and characterization of aptamer. Whole procedure consist from the following steps: isolation and immobilization of NF-M, preparation of compounds for depletion of oligonucleotides from library of oligonucleotides that might unspecifically bind to NF-M, preparation of oligonucleotide library for screening, selection and amplification of oligonucleotides that bind to NF-M, binding assay with labelled oligonucleotides, cloning and sequencing of selected oligonucleotides and functional chracterization of synthetic aptamers.
