MALDI-TOF MS quantification is hampered by the poor reproducibility and molecular-mass and compositional discrimination. The use of an internal standard increases reproducibility and allows a calibration curve to be constructed. With synthetic polymers, the method requires a polymer of the same molecular mass distribution and composition as the analyte. When the content of the precursor polymer in a modified polymer is sought, the pure precursor is usually available and the modified polymer can serve as an internal standard. The method has to be combined with that of standard addition and the precursor polymer added directly to the sample. The experiments with simulated modified polymers revealed the signal-intensity ratio not proportional to the concentrations ratio. Thus, the basic assumption of the standard-addition method is not met. Therefore, the multi-point combined internal-standard standard-addition method based on the power-type calibration curve was developed and verified.
