    AnnotationASLV family retroviruses contain 5 highly related envelope subgroups (A-E). Susceptibility or resistance of cells to infection by these ASLVs is given by 3 loci in chicken. The tvb gene encodes subgroup B, D, E ASLV receptors. Wild-type TvbS1 receptor confers susceptibility to subgroup B, D, E ASLVs. In this study the genetic defect underlying altered susceptibility of inbred chicken line M to infection by ASLV(B), (D), (E), C125S substitution in tvb-encoded TvbS1 receptor in this line, was identified. We show that this substitution significantly reduces susceptibility of line M cells to infection by ASLV(B) and (D) and virtually eliminates susceptibility to ASLV(E) infection in cultured cells and the incidence and growth of ASV-induced sarcomas in chickens. It also significantly reduces binding affinity of TvbS1 receptor for subgroup B, D, E ASLV envelope glycoproteins. These results are first demonstration of possible role of the Cys-rich domain 3 in the function of Tvb receptors.
