To test the hypothesis that changes in intrinsic optical signals (IOS) correspond to activity-related cell swelling, we simultaneously measurd changes in light transmittance (LT) and extracellular space diffusion parameters (ACS-DP) evoked by a 45 min application of hypotonic solutions or elevated K+ in spinal cord slices of 910-day-old-rats. We conclude that an LT increase is not directly related to cell swelling, but ratherto fine structural changes corresponding to K+-evoked neuronal activity.
