The development of Bioartificial Liver Assist Devices (BLADs), intended for the therapy of chronic and fulminant liver failure, is one of the most important tasks in the area of tissue engineering. The primary objective of our research was to examine the interactions of cell cultures (hepatocytes, bone marrow mesenchynal stromal cell mixed cultures) with nanofiber-based materials, which can be utilized as carriers for high density cultures, and to test the possibility of creating a polarized hepatocyte culture on a nanofiber layer., separating two compartments with a different composition of the cultivation medium (simulating the capillary and bile compartments of the hepatic tissue). The results reported demonstrate that nonwoven nanofiber layers are a suitable biocompatible support for functional, hepatocyte cultures and can be utilized in a laboratory bioreactor; mesenchymal stromal cells from bone marrow may be utilized in order to enhance hepatocyte functionality on the nanofiber layers.
