The purpose of the study was to compare predifferentiated ASCs (pASCs) with naïve ASCs in vitro and after transplantation into rats with a balloon-induced spinal cord compression lesion. ASC spheres were induced prior to transplantation, then pASCs or ASCs were injected intraspinally. Behavioral, histological and electrophysiological methods were used to assess the cells' fate and the rats' functional outcome. The predifferentiation of ASCs facilitated the protection of denuded axons; however, functional improvement was comparable in both groups, indicating that repair was induced mainly through paracrine mechanisms.
