Recognition and the binding of spermatozoa to the zona pellucida (ZP) is a crucial step in the fertilization process. Despite longstanding research efforts to identify the sperm proteins that recognize ZP receptors, their precise determination still remains questionable in various mammalian species. Sperm proteins of ejaculated and in vitro capacitated boar sperms were isolated by agents as follows: Triton X-100, Triton X-114, acetic acid, SDS, n-octyl-β-D-glucopyranosid, extraction by freezing-thawing and characterized by 1D, 2D electrophoreses glycoprotein staining and substrate zymographic methods. Results have shown the quantitative and qualitative differences in 1D, 2D protein and glycoprotein profiles depending upon the isolation protocol. Differences were observed between protein profiles of ejaculated and capacitated sperms as well. Zymograms supported the prediction that using various isolation protocols gives various profiles of enzymatically active molecules.
