So far, transgenic RNAi in mouse oocytes has been using long dsRNA (over 500 bp long). Although it worked well in the oocyte, this technology cannot be used in somatic cells and it requires a considerable amount of work. Therefore, we tested a universal shRNA system placed in a retrovirus-compatible vector. Our results show that the shRNA technology does not bring any advantage in the amount of labor, time and cost and, furthermore, this particular type of vector is efficiently epigenetically silenced in transgenic animals.
