    Food intake is activated by hypothalamic orexigenic neuropeptide Y (NPY) controlled by leptin and ghrelin. Rat adjuvant arthritis (AA), and also human rheumatoid arthritis, is associated with cachexia. In our study, we evaluated NPY expression in hypothalamic arcuate nucleus (nARC) under AA-induced changes in leptin, ghrelin and adiponectin. Anorexia is centrally induced by IL-1β whose expression in the nARC was studied as well. In AA rats, decreased appetite, body mass, and epididymal fat positively correlated with reduced circulating and epididymal fat leptin and adiponectin. Plasma ghrelin was increased. In nARC, mRNA for leptin and ghrelin receptors, NPY, and IL-1beta were over-expressed in AA rats. However, circulating and spleen IL-1beta was unaltered in AA rats. During AA, despite of over-expression of orexigenic NPY in nARC, activated expression of IL-1beta in nARC kept AA-induced anorexia in progress. Lowered adiponectin might contribute to this anorexia.
