We have cloned, sequenced and characterized an macromolecular protease inhibitor of α2-macroglobulin family from the hard tick Ixodes ricinus named IrAM. Using the tool of RNA interference we have demonstrated that IrAM is an important constituent of tick innate immunity since it is involved in the phagocytosis of the pathogen Chryseobacterium indologenes by tick hemocytes. This is the first evidence showing that a cellular immune response in invertebrates is linked with interaction of an plasma α2-macroglobulin with protease derived from invading pathogen.
