The maize beta-glucosidase Zm-p60.1 is important for the regulation of plant development through its role in the targeted release of cytokinins from cytokinin-O-glucosides, their inactive storage forms. Enzyme kinetics studies using these scarce substrates close to physiological concentrations are difficult. We developed a glucose assay using a system comprising three enzymes - beta-glucosidase, glucose oxidase and horseradish peroxidase, with Amplex Ultra Red reagent.
Kukuřičná beta-glukozidáza Zm-p60.1 hraje důležitou roli v regulaci vývoje rostlin vzhledem k její roli v cíleném uvolňování cytokininů z jejich O-glukozidů, které jsou jejich neaktivními formami. Studium enzymové kinetiky s použitím těchto vzácných substrátů v blízkosti jejich fyziologických koncentrací je obtížné. Vyvinuli jsme postup pro stanovení glukózy s využitím systému zahrnujícího tři enzymy - beta-glukozidázu, glukózaoxidázu a křenovou peroxidázu, s reagencií Amplex Ultra Red.
