In Tetrahymena thermophila and Paramecium caudatum, immunofluorescence microscopy with the monoclonal antibody TU-06, directed against b-tubulin, revealed distinct staining of this cortical region alone, while the cilia and other microtubular structures were unstained. Double-label immunofluorescence with antibodies against g-tubulin, detyrosinated a-tubulin, and centrin showed that the TU-06 epitope is localized outside the basal body region. This was also confirmed by immunogold electron microscopy of thin sections. Proteolytic digestion of porcine brain b-tubulin combined with a peptide scan of immobilized, overlapping peptides disclosed that the epitope was in the b-tubulin region b81–95, a region which is phylogenetically highly conserved. The limited distribution of the epitope could rather reflect the dependence of TU-06 epitope exposition on conformations of tubulin molecules in microtubule arrangements or on differential masking by interacting proteins.
V nálevnících rodu Tetrahymena a Paramecium (trepka) byla popsána neobvyklá lokalizace b-tubulinového epitopu. Tento epitop se nachází v evolučně konzervované oblasti b-tubulinu (81-95) a je rozpoznáván myší monoklonální protilátkou TU-06. Imunofluorescenční i elektronová mikroskopie ukázaly přítomnost TU-06 epitopu výlučně v kortikální oblasti buněk, tj. pod plazmatickou membránou, ale nikoli ve struktuře bazálních tělísek. Takto omezená distribuce b-tubulinového epitopu může odrážet odlišnou dostupnost epitopu na různých strukturách v závislosti na konformaci tubulinových molekul nebo vlivem maskování epitopu pomoci interagujících proteinů.
