Cell staining with Propidium iodide offered an effective tool for monitoring the progress of the cell lysis. Complete lysis in the microplate was reached with 0.01% (v/v) of the Triton X-100 after 10 minutes of passive detergent diffusion while 0.1% (v/v) of detergent was used for cell lysis in the PDMS droplet-based microfluidic chip. The proposed approach can be advantageously used for further cell manipulation and cell storage as well.
