    In this work, we present design, synthesis, and application of new fluorescent tags providing rapid labeling kinetics, high fluorescence quantum yields, and significantly enhanced detectability by MS. The new fluorescent tags based on pyrene, coumarin and/or BODIPY derivatives with various substituents and modifiable linkers were synthetized and the desired modification allowed to tune the fluorescence and MS properties (preferred wavelength and quantum yield, charge). A detailed comparison of the mechanisms of labeling, reductive amination and hydrazone formation chemistries, showed that the labeling yield obtained by hydrazone formation chemistry is almost one order of magnitude higher compared to the reductive amination approach. Furthermore, the undesired formation of multiple products of N-linked glycan labeling by reductive amination was detected when positively charged labels such as aliphatic/aromatic amines containing a quaternary ammonium group were attached. Labeling with different synthetized tags as well as their comparison with the commercially available labels will be demonstrated by analysis maltooligosaccharides and N-linked glycans by CE/LIF and CE-MS.
