    Reversed-phase UHPLC/MS is a widely used lipidomic strategy that allows the separation of the lipid species based on their equivalent carbon number (ECN - the number of carbons minus two times the number of double bonds of all fatty acids). Lipid classes as well as fatty acid compositions are determined using fragments in MSn spectra. Retention time has to correspond with identifying lipid species, so the knowledge of retention dependences significantly enhances the certainty of identification. Increasing chromatographic peak resolution allows identifying more lipid species, and short run times are essential for analyzing large sets of samples in lipidomics. Reducing the slope of the gradient is not a suitable solution for increasing resolution, so selecting a suitable stationary phase is critical. In this study, four C18 columns with sub-2-micrometer particles were compared using a mixture of 71 lipid standards from 16 lipid classes.
