Polyomavirus ATPase, DNA helicase, and DNA binding activities of polyomavirus large tumor antigens (Tags) are diminished by low concentrations of alkoxyalkyl-esters of acyclic nucleotide phosphonates (ANPs). Inhibition of viral DNA replication is mediated by inhibiting ssDNA and dsDNA binding activities of Tags. The alkoxyalkyl-esters of ANPs were found effective in vitro without undergoing further conversions.
