Random gene trap mutagenesis is a powerful technique to rapidly generate a library of mutated genes while also providing a reporter for their respective expression. One problem is that efficient mutagenesis requires insertion of the vector at the beginning of a gene, while an informative subcellular reporter requires insertion at the end of a gene. Here we introduce a novel gene trap approach that overcomes this issue, creating a multifunctional allele that permits both effective mutagenesis and endogenous protein tagging.
