Intestinal organoid cultures represent a suitable model for studying the organisation and homeostasis of the intestinal epithelium in vitro. Here we present a step-by-step protocol for establishing and propagating organoid cultures from the freshly isolated mouse small intestine and colon. We provide detailed information related to materials and workflow based on the original procedure that has been modified according to our experience.
