The present invention relates to an insulin derivative of formula I, consisting of des(B23-B30)octapeptide-insulin (DOI) and a modified octapeptide, which form an amide bond between the carboxyl group of C-terminal ArgB22 in the DOI molecule and the amino group of N-terminal GlyB23 in the molecule of the modified octapeptide comprising a cyclic structure. The des(B23-B30)octapeptide-insulin is human insulin without the C-terminal octapeptide B23-30 and the modified octapeptide imitates the B23-B30 sequence of human insulin. The insulin derivative of formula I is suitable for use as an anti-diabetic agent with a fast onset of action and low risk of cancer propagation.
