Studies in dynamic changes in protein translation require specialized methods. In this paper, we examine changes in newly-synthesized proteins in response to ischemia and reperfusion using the isolated perfused mouse heart coupled with polysome profiling. We present the methods that were used to analyze the distribution of proteins, mRNAs, and miRNAs in the fractions after sucrose gradient centrifugation. Specifically, in the high-efficiency fractions (numerous ribosomes attached to mRNA), low-efficiency fractions (fewer ribosomes attached), and the non-translating fractions.
