Using DNA bisulfite treatment, reducible cytosines were transformed into nonreducible uracil residues, strongly decreasing square wave voltammetric C reduction peaks of unmethylated DNA. On the other hand, 5-methylcytosines were not transformed and the reduction peaks of methylated DNA remained relatively unchanged. By combining DNA bisulfite treatment with square wave voltammetry, DNA methylation can be determined quantitatively at nanomolar and subnanomolar ODN concentrations.
