We used reversed phase LC under various experimental conditions to optimize preseparation of protein complex mixtures of cardiac myocyte secretomes and plasma prior to mass spectrometry identification. The effect of hypoxia on cardiac myocytes and fibroblasts was tested as well. The analysis of proteins secreted by various cells is an alternative approach to the direct analysis of plasma for discovery of potential protein biomarkers and therapeutic targets.
