    Leishmanias are causative agents of leishmaniases and an important biological model for study host-pathogen interaction. Many methods of their cultivation on biphasic and liquid media are available. Biphasic media are superior for initial isolation of parasites and obtaining high promastigote infectivity; liquid media are more suitable for large-scale experiments. Our study adapted and optimized cultivation of Leishmania promastigotes on biphasic SNB-9 (saline–neopeptone–blood 9) originally developed for Trypanosoma cultivation, combining advantages of biphasic and liquid media. We demonstrate that SNB-9 considerably surpasses Schneider's Insect Medium in in vitro infectivity of the parasites and show that the ratio of apoptotic parasites important for the infectivity of the inoculum is higher in SNB-9 culture than in Schneider's Insect Medium culture. Cultivation of Leishmania on SNB-9 thus reliably yields highly infective promastigotes suitable for experimental infection.
