Modern electron microscopy research demands appropriate stabilization of biological samples securing the best ultrastructural and antigenic preservation of molecular complexes. High-pressure freezing (HPF) of cells appears to be the method of primary choice for examining macromolecular assemblies in situ in close-to-native state leading to a better understanding of the functions of complex cellular structures [1]. Here we present results of our experiments aiming to improve the quality of biological samples in order to achieve an optimal preservation of both the ultrastructure and antigens before the subsequent immunogold labeling on ultrathin sections. All the described evaluations concern combinations of HPF and automated freeze substitution (FS) followed by resin embedment.
