Targeting of proteins to DNA double strand breaks is highly organized hierarchical process, which depends on posttranslational modifications such as phosphorylation, ubiquitylation and sumoylation. This short review summarizes newly discovered role of protein HERC2, which is targeted to DNA double strand breaks by ATM kinase mediated phosphorylation, where it serves as adaptor protein for ubiquitylation enzymes E3 ligases RNF8 and RNF168 and specifity factor for E2 ligase UBC13. Possible relationship of HERC2 genetic alterations to some human diseases is also discussed.
