Polymer-drug conjugates represent a novel generation of polymer therapeutics enabling site-specific drug action what predestines them predominantly for tumor therapy. In this review a design, synthesis and physico-chemical and biological properties of water-soluble synthetic polymer-drug conjugates including principles of tumor-specific passive and tumor cell-specific active targeting, controlled drug release and elimination of the polymer drug carrier from organism is discussed and potential of polymer-drug conjugates for tumor treatment is evaluated.
