V-taper probes were prepared by tapering a stack of two polymer-clad-silica (PCS) fibers to tips with diameters in a range from 8 to 50 um. A fluorescence transducer 2’,7’-Bis(2- carbonylethyl)-5(6)-carboxyfluorescein (BCECF) was immobilized onto the end-face of the cut V-tapers by using a thin tetraethoxysilane layer. By using a PCS V-taper probe with a tip diameter of 44 μm excited by a multimode pigtailed source, extracellular pH was determined in real cell samples with the resolution of 0.2. Plant-cell walls were successfully penetrated with these probes.
