n DESI, an electrospray emitter aims at surface of interest and produces primary charged droplets that impact the surface and pick up the analyte (according to prevailing droplet pick up mechanism) that is transported into a mass spectrometer and ionized by processes similar to those that take place in the standard ESI. DESI is used mainly to analyze small to medium size polar molecules (e.g. drug metabolites, organic compounds, phospholipids, peptides) often in very complex environments and matrices (e.g. tissues, cell cultures)
