Lincomycin and its derivatives are potent antibiotics used in human and veterinary medicine. Lincomycin molecule is composed of two subunits, methylthiolincosamide (MTL) and methylproline linked by an amide bond. The gene cluster of Streptomyces lincolnensis controlling lincomycin biosynthesis contains 29 ORFs with putative biosynthetic/regulatory (lmb) and resistance (lmr) functions. Six ORFs (lmbA, B1, B2, W, X, Y) were found to have their counterparts among genes controlling biosynthesis of anthramycin sharing with lincomycin the initial reactions of biosynthesis of proline derivatives. Functions of LmbB1 and LmbB2 proteins were demonstrated in vitro; by gene inactivation analysis lmbA, X, W genes were confirmed to be included in propylproline biosynthesis
