To assess the role of distict protein domains we created the mutants of S. pneumoniae expressing the epitope labeled full-length and truncated forms of StkP. The mutants were prepared by allelic exchange. We propose that the deletion of C-terminal part of StkP including the transmembrane domain leads to the instability of the truncated protein. Anchoring of the kinase in the membrane resulted in the expression of stable protein with an autophosphorylation activity. We provide the evidence that both the transmembrane and the C-terminal domains possess a capacity to form dimers
Ve snaze určit roli jednotlivých domén jsme pomocí alelické výměny připravili mutantní kmeny S. pneumoniae exprimující epitopem (oligohistidin) značenou nezkrácenou a epitopem značené zkrácené formy StkP. Delece C-terminální části včetně transmembránové domény se projevila nestabilitou zkráceného proteinu. Ukotvení kinázové domény do membrány vedlo k expresi stabilního proteinu s autofosforylační aktivitou. Prokázali jsme, že jak transmembránová, tak i C-terminální doména se podílí na dimerizaci proteinu
