We use volume reconstruction as a technique for visualization of biological tissue specimens which are greater than the field of view of a confocal laser scanning microscope. Prior to the volume reconstruction, large specimens are cut into thin physical slices. The first step of volume reconstruction is acquisition of sets of digital volume images (spatial tiles which overlap) from all studied physical slices. The second step is horizontal merging of overlapping spatial tiles of the same physical slice (stitching). The third reconstruction step represents vertical merging of digital volumes of successive physical slices of the specimen. We applied a curve-driven elastic registration algorithm which models deformations by using B-splines. The resulting large digital volumes are visualized using a VolumePro hardware board that offers real-time 3D volume rendering. In this paper we show reconstruction of a laboratory rat oesophagus

Článek popisuje objemovou rekonstrukci a vizualizaci biologických tkáňových vzorku s použitím konfokální mikroskopie, jejichž velikost je větší než zorné pole a hloubka proniku konfokálního mikroskopu. Je využívána registrace obrazů s použitím Euklidovských transformací a elastických deformací modelovaných pomocí B-splinů
