In the presented thesis, optical biosensor platform for high-throughput label-free monitoring of biomolecular interactions is reported. This platform is based on angular spectroscopy of surface plasmons on an array of metallic diffraction gratings. Based on performed theoretical analysis, prototype SPR sensor device including an SPR sensor chip with a two-dimensional array of sensing channels, fluidic system and SPR chip reader was developed and its main performance characteristics were determined.
V rámci této disertační práce byl vyvinut nový optický biosensor pro vysoce přesnou a paralelizovanou analýzu biomolekulárních interakcí. Tento sensor je založen na dvoudimenzionální matici kovových diffrakčních mřížek a angulární spektroskopii povrchových plazmonů. Byla provedena teoretická analýza senzoru, na základě které byl realizován a otestován prototyp zařízení zahrnující senzorový čip s dvoudimenzionální maticí senzorových kanálů, fluidický systém a optický systém.
