The cultivation protocol of Norway spruce embryogenic cultures was elaborated. The cultivation protocol consists of all phases of somatic embryogenesis, e.g. the induction of embryogenic culture, long-term maintenance of embryogenic cultures on proliferation medium, the development and maturation of somatic embryos, their desiccation and subsequent germination. The process of cryopreservation of embryogenic cultures, enabling long-term storage of tissues before their further cultivation, is described in the protocol. The vital staining of embryogenic cultures, the preparation of microscopic sections and their staining, and the use of computer image analysis are also included.

Byl vypracován kultivační protokol pro embryogenní kultury smrku ztepilého (Picea abies). Protokol zahrnuje všechny fáze somatické embryogenese, tj. indukci embryogenní kultury, dlouhodobé udržování kultur na proliferačním médiu, vývoj a maturaci somatických embryí, jejich desikaci a následné klíčení. Součástí je i postup umožňující uchovávat embryogenní kultury zmražené v tekutém dusíku s možností kultury popsaným způsobem rozmrazit a dále kultivovat. V protokolu je pojednáno i vitální barvení embryogenních kultur, barvení a příprava mikroskopických řezů a jejich hodnocení pomocí počítačové analýzy obrazu.
