Using of FITC-VAD-FMK (or other FLICAs) for detection of caspase-like activity leads mostly to artefactual staining of cells picked up from ageing Saccharomyces cerevisiae colonies. On the contrary, (Asp)2-Rhodamine (successfully used as a caspase substrate in mammalian cells) appears to be suitable substrate for detection of active ASPases (proteases able to cleave substrate after Asp residue) in yeast cells
