AnotaceThe aim of our study was to clarify if relapses in clubfoot therapy are associated with altered angiogenesis and to suggest possible regulatory pathways of this pathological process. This hypothesis was confirmed by the detection of a significant increase of microvessel and arteriole density in the contracted medial-side of the relapsed idiopathic clubfoot (determining the functional heterogeneity of endothelial cells would be beyond the scope of this article). We described the methodology of the quantification of microvascular and arteriole density. We counted all the vessels with CD31 positive endothelial cells without muscular wall in the tissue sample. Arterioles were identified by the detection of their internal elastic lamina. We also counted all arterioles in the tissue sample, which were completely surrounded by dense irregular connective tissue. Tissue samples for the purposes of this study were taken from relapsed patients who had undergone a surgical procedure after an unsuccessful casting treatment. Although it is one of the most common birth deformities and occurs in 1 out of every 1000 live births, indication for surgery treatment is very rare. For future research, we would like to enlarge our sample group to include tissue samples from new patients, which would make our results even more significant.
