Here we describe the synthesis, physico-chemical characteristics and detailed study of degradability of graft, diblock or multiblock HMW polymer carriers based on HPMA copolymers or PEG designed for delivery of drugs by passive tumor targeting. The components of the polymer carriers, grafts, main chains and blocks with molecular weight below 50 000, are linked via spacers containing disulfide groups susceptible to reductive degradation. We have performed in vitro degradation of the polymer carriers in a buffer containing glutathione and compared with that in the presence of EL4 T-lymphoma cancer cells.
