In continuation of this work we have been exploring small molecules with permanent positive charge as cationic tags. Oligosaccharides and glycans were labeled by reductive amination and the resulting derivatized saccharides were analyzed by capillary electrophoresis coupled to contactless conductivity detection and electrospray ionization mass spectrometry. The quaternary ammonium based tags assured migration of the oligosaccharide species as cations with electrophoretic mobility independent of the pH of the background electrolyte. For transient isotachophoretic preconcentration, the sample was complemented with ammonium ions and injected as a long zone occupying up to 70% of the separation capillary length. The labeled species, injected in the nanomolar concentrations, were focused into narrow zones by transient isotachophoresis followed by the separation in capillary electrophoretic mode.
